My sister-in-law has an unlocked iPhone that she's been using with T-Mobile. My wife and I both have 3GS's with AT&T. We want to add her to our family plan so what's the best way for her to relock her phone so she can use AT&T with it again??

She can restore the phone....as new or from backup. Either one will erase the jailbreak/unlock. She can then upgrade to the latest firmware. Then go to ATT and activate phone.
Edit: probably have to go to ATT after the restore as the sim will be invalid. She can upgrade firmware once phone is reactivated.

Eh, I would restore to new since back up can leave traces of the jailbreak on her phone. The only pain is that you have to sync everything again.

You also dont ahve to "re-lock" the iPhone, it will work with any carrier - including ATT

It doesnt need to be relocked she just needs to change the sim card to an AT&T sim card

If AT&T reps aren't going to have to check the iPhone out to activate it or whatever, then yeah, no need to relock it.

cancel the account at t-mobile
add another line to your account at at&t
insert sim card
...no need to unlock and the best thing is that if you guys choose to change it back, you don't have to unlock again. once you unlock, you shouldn't relock it for future cases.

Cool. Thanks all. I didn't realize it was as simple as restoring as new...
